Chlorogenic Acid, Rutin Content and Physiological Activities of Cudrania tricuspidata Fruit Extract

Abstract
This study was conducted to evaluate physiological activity like antioxidant, antibacterial, anti-inflammatory, and digestive enzyme activity in water extract of Cudrania tricuspidata fruit(CTW). The IC50 of DPPH and ABTS radical scavenging activities of CTW were 6.79 ¡¾ 0.09 mg/mL, 8.69 ¡¾ 0.05 g/mL, and total polyphenol and total flavonoid contents were 99.62 ¡¾ 1.11 mgGAE/g, 50.3 ¡¾ 2.4 mgQE/g, respectively. The 400 ¥ìg/mL of CTW performed more than 41.3 ¡¾ 0.5% protective effects against oxidative stress in HepG2 cell lines. Antibacterial activity increased as the dose concentration of CTW increased and the MIC was 100 mg/mL for L. monocytogenes, S. Typhimurium, and 120 mg/mL for H. pylori. CTW did not show cytotoxicity in RAW 264.7 cells and Caco-2 cells, and inhibited NO production in concentration dependent. NO production was reduced by 71.3 ¡¾ 1.3% in RAW 264.7 cells and 67.7 ¡¾ 1.8% in Caco-2 cell. Similarly, the production of inflammatory cytokines (TNF-¥á, IL-1¥â, and IL-6) was inhibited in a concentration dependent manner. Furthermore, amylase and protease enzyme activity was observed in the CTW and the enzyme activity was increased according to the concentration of the extract. CTW contained 13.12 ¡¾ 0.11 mg/g of chlorogenic acid and 1.23¡¾0.01 mg/g of rutin. The results indicate that it can be used as basic data for the development of health food.
